I’ve set a minimum buy for my store. Bricklink can’t account for the zones used
by USPS Ground Advantage. I added the minimum buy to avoid a situation where
the actual shipping costs more than the price of the set and the paid shipping
combined. $10 is higher than what many other stores do, so I’m wondering if adding
manual invoicing for small purchases could be worthwhile.

I think for your store and what you're selling , $10 min is quite ok.
I recently switched from a $2 minimum to a $5 minimum. I didn't mind the
$2 sales, but it was a lot of work to break even or lose money even. Even so,
it has not negatively affected my sales.

Yes, minimum buy will deter purchases. The whole point of a minimum buy is to
deter purchases that are less than what you want to sell for. Will they deter
purchases that are over your minimum amount? No. Why would they?
